Interleukin-6 (IL-6), a 22–28 kDa glycoprotein composed of 187 amino acids in mice, plays a critical role in inflammation, immune regulation, hematopoiesis, and disease progression. It is secreted by various cell types, including macrophages,
dendritic cells, fibroblasts, endothelial cells, and T cells, in response to infections, tissue damage, and inflammatory stimuli. Elevated IL-6 levels are associated with autoimmune
diseases, metabolic disorders, cardiovascular diseases, and cancer progression in murine models.

As a key cytokine in innate and adaptive immunity, IL-6 influences B-cell differentiation, T-cell activation, and acute phase responses. It signals through a heterodimeric receptor complex composed of IL-6R and glycoprotein 130 (gp130), triggering downstream pathways such as JAK/STAT3 and MAPK that regulate immune responses, hematopoiesis, and tissue homeostasis.

Unlike constitutively expressed cytokines, IL-6 is inducible and responds dynamically to pathogens, cytokines, and stress signals. Its pleiotropic effects range from promoting inflammation to tissue regeneration, depending on the biological context. Dysregulated IL-6 production is implicated in chronic inflammatory diseases and cancer, making it a key biomarker in murine disease models. Measuring IL-6 levels in mouse serum and plasma provides valuable insights into disease mechanisms and therapeutic interventions in preclinical research.
